1. 研究氧化壓力對細胞老化及分化之影響

      在物種演化過程上,原始生命型態開始使用氧氣呼吸,即遇上兩難局面。一方面藉有氧呼吸有效產生能量;另一方面氧氣可產生活性氧分子。活性氧分子帶有高度活性,可氧化並破壞細胞內巨分子,而改變細胞生理狀態。其中巨分子氧化損傷累積即可使細胞生長停滯,進入老化狀態。我們使用帶有抗氧化酶遺傳缺陷細胞作模式,研究氧化損傷造成老化所伴隨的基因表現變化。同時,我們使用RNA干擾技術,抑制不同抗氧化物基因表現。進而研究氧化壓力對細胞生理及相關蛋白質體及轉錄體。

2. 研究粒線體與氧化壓力在癌症發生之角色

      粒線體被認為是活性氧分子主要來源;亦是能量產生及其它生理過程如細胞淍亡之調控者。癌細胞與正常細胞的代謝有極大差異。我們的初步研究結果顯示:粒線體狀態與癌細胞生長密不可分。目前,我們使用分子技術及動物模式,探討其機轉。同時,我們使用RNA干擾技術,研究粒線體及氧化壓力對癌症發生之角色。

3. 研究氧化壓力與感染性疾病之關係

      宿主因素影響病原體的感染性及致病性。氧化還原態為宿主因素之一。其具體角色仍未完全明瞭。我們的初步研究結果顯示:宿主氧化還原態若偏向氧化態時,腸病毒的複製及所引至之細胞病變效應即加劇。粒線體狀態在病毒感染時呈異常。目前,我們使用分子技術及動物模式,探討其機轉。
